Introduction:

Introduction: Triple negative breast cancer (TNBC) presents a formidable challenge in oncology due to its aggressive nature and limited treatment options. Current drug screening models, predominantly based on 2D cell cultures, often fail to replicate the complex microenvironment and chemotherapy resistance commonly observed in patients, resulting in suboptimal drug development outcomes. This study investigates the potential of using thermal inkjet bio-printed (TIB) cells as a novel model to enhance drug screening by evaluating paclitaxel and cyclophosphamide cytotoxicity in bio printed cells. This will help to develop more resistance cell lines that can be used to identify drugs that are more efficient to treat patients with chemotherapy resistant TNBC.

Methods:

Methods: To assess the chemotherapy efficacy of Paclitaxel and cyclophosphamide that are commonly systematic drugs used to treat TNBC, we evaluated the cytotoxicity of these drugs against thermal inkjet bio-printed MDA MB 231 cells versus manually seeded MDA MB231 cells. 2,000 bio printed or non bio printed cells were seeded into a 96 well plate. Eight dilutions of paclitaxel or paclitaxel in combination with cyclophosphamide were used in triplicates to identify the concentration need it to kill 50% of the cells (IC50). After a two-day incubation with the chemotherapy, 1uM of Hoechst and propidium iodide were added into each well and incubated for one hour at 37 degrees. Cells were counted using a 1x Pico analyzer and percentage of death cells were used to identify the IC50 of each condition in both cell type

Results, Conclusions, and Discussions: RESULTS/

Discussion: Chemotherapy resistance was observed on bio printed cells compared to manually seeded cells. In manually seeded cells, there was not an observed increase in cytotoxicity during co-treatment with cyclophosphamide; however, bioprinted cells showed a slide increase in cytotoxicity during co-treatment. Comparing manually seeded cells and bio printed cells; bioprinted cells were more resistant to paclitaxel and cyclophosphamide co-treatment. The IC50 observed in manually seeded cells was 15.62nM in comparison to bioprinted cell with an IC50 of 115.7nM. TIB cells exhibited increased resistance to Paclitaxel and Cyclophosphamide treatment, suggesting their potential in developing more resilient models for drug testing in TNBC. Thermal inkjet bio-printing of MDA MB231 cells enhances their chemotherapy resistance, positioning them as promising candidates for advanced drug screening models in TNBC. These findings underscore the utility of TIB technology in recapitulating in vivo cell behavior and addressing the limitations of conventional 2D cultures.

Conclusion: This study addresses the transformative potential of TIB technology in cancer research, particularly in the development of robust models for studying drug resistance mechanisms in TNBC. Further exploration of TIB-induced gene expression changes and long-term culture viability is crucial to fully harness its applications in personalized medicine and drug discovery. Implementing TIB-derived models holds promise for accelerating the identification of effective therapies against chemotherapy-resistant TNBC, ultimately improving clinical outcomes for patients.
