Poster Y14 - Magneto-mechanical actuation of magnetic nanoparticles for treatment of nascent surgical adhesions

Introduction:  

Introduction: Surgical adhesions are the most frequent complication of abdominal surgery and are the leading cause of secondary infertility in women. A promising approach to treat surgical adhesions is the mechanical disruption of the fibrin gel matrix that comprises the primary structural component of adhesions. Remote mechanical disruption of nascent surgical adhesions may be possible with superparamagnetic iron oxide nanoparticles (SPIONs) in a low frequency dynamic magnetic field (LFDMF). In contrast to high frequency (>1kHz) alternating magnetic fields that induce heat generation of SPIONs, LFDMFs can physically rotate SPIONs in the direction of an applied magnetic field in a process known as magneto-mechanical actuation.

Methods:  

Methods: Our LFDMF device is constructed based on a nested Helmholtz coil configuration. Each coil pair is wound in series using 18 American Wire Gauge (AWG) enameled copper wire and connected to a high current H-bridge that changes the direction of current flowing through the coils at a frequency controlled by the user via Arduino software. The magnetic field strength of the LFDMF device is measured using a Hall probe. The percentage of transmitted light traveling through a polystyrene cuvette filled with 1mg/mL SPION in deionized water or 80% glycerol in water solution subjected to LFDMF treatment was measured via OceanOptics USB2000+ spectrometer.

Results, Conclusions, and Discussions: 

Results: In the present study, we focus on the design and construction of a low frequency dynamic magnetic field (LFDMF) device capable of producing magnetic field strengths up to 24mT at frequencies between 1-100Hz. Aggregation and rotation of SPIONs in water and viscous fluid is observed via spectrophotometry. The change in percentage of transmitted light of SPIONs suspended in deionized water solution subjected to LFDMF treatment is 20.5%. For SPIONs in viscous glycerol/water solutions, the change in percentage of transmitted light of SPIONs is 5.6%.

Conclusion: We have developed a scalable dynamic field device and demonstrated magneto-actuation of SPIONs. The change in transmitted light when an applied magnetic field is pulsed in parallel and perpendicular directions to the light source indicates aggregation and rotation of SPIONs in viscous fluid. Overall, this study indicates potential for magneto-mechanical disruption of nascent surgical adhesions using SPIONs in a LFDMF.
