Introduction: Optical coherence tomography (OCT) is an optical imaging method using backscattered light to generate depth-resolved, cross-sectional images of tissue. OCT is widely used to assess retinal diseases in clinics, and is also used in numerous research studies to examine tissue characteristics in skin and ear. The goal of this research is to build a portable optical imaging system that can be used for both retinal and ear assessment using spectral-domain OCT (SD-OCT).

Materials and

Methods: Our SD-OCT system was custom-built using a superluminescent diode (Superlum Diodes) centered at 840 nm with a bandwidth of 50 nm (full-width-half-maximum) and a high-speed spectrometer (max line-scan rate of 250 kHz, Wasatch Photonics). A compact galvanometer scanner (ScannerMax) was used to obtain B-scans and volumetric datasets. A data acquisition card (PCIe-6351, National Instruments) was utilized to trigger and generate analog signals for the scanner. A 2 x 2 fiber coupler divided the light into a 75:25 coupling ratio to reference and sample arm, respectively. A dichroic mirror was used to provide a simultaneous USB camera view (Ximea). An achromatic doublet was used in the sample arm for ear imaging, and additional achromatic doublet was inserted for retinal imaging. A linear stage in the reference arm adjusted different optical path lengths between retinal and ear imaging.

Results, Conclusions, and Discussions: Figure 1 describes the schematics of our imaging system. For triggering and acquisition of OCT signals, we implemented an open-source OCTSharp software written in C#. The incident power on the sample was 2.8 mW, but it was kept below 2 mW for retinal imaging. Multiple handheld probe designs and materials were evaluated for clinical feasibility, such as size, weight, color, and durability. Benefits and challenges from implementing the open-source OCTSharp software in a custom-built OCT system will be discussed. A single, portable device capable of both retinal and ear imaging will facilitate multiple translational imaging studies. Future work includes continued optimization and characterization of the OCT system and probe, implementation of computer-controlled reference arm path length changes, and demonstration on healthy volunteers and patients in a clinical setting.
